Description

Description

One Shot LB Agar Plates are pre-poured plates containing Luria-Bertani (LB) agar medium that are available with or without antibiotics. LB agar is a solid bacterial growth medium used in molecular biology studies for the cultivation and maintenance of Escherichia coli strains. Autoclaved glass beads are included to aid in plating.

LB agar is a popular, nutrient-rich medium made from casein peptone, yeast extract, sodium chloride, and agar. Casein peptone or tryptone is an enzymatic hydrolysate of casein. It supplies nitrogen, carbon, and amino acids. Yeast extract provides trace elements, vitamins (including B group vitamins), amino acids, and peptides. Sodium chloride supplies sodium cations for membrane transport and maintains osmotic balance. Agar is a solidifying agent of the growth medium.

One Shot LB Agar Plate composition is based on the Jeffrey H. Miller formulation and contains twice as much sodium chloride as the Lennox formulation. The inclusion of autoclaved glass beads allows for simple plate inoculation without the need for additional loops or spreaders.

As E. coli is unable to digest agar but can absorb nutrition from the LB medium, single colonies can be easily isolated and selected. On plates with a selective antibiotic only transformed E. coli with resistance can grow.

One Shot LB Agar Plate formulation

10 g/L casein peptone, 5 g/L yeast extract, 10 g/L NaCl, 15 g/L agar

Benefits of One Shot LB Agar Plates

  • Read-to-use format conveniently packaged in two sleeves of 10 plates
  • Long-shelf life: 3.5 months without antibiotic and 3 months with antibiotic
  • Autoclaved glass plating beads provided for easy cell spreading

One Shot LB Agar Plates are a good choice for use with our Gateway, TOPO, and Gibson Assembly cloning kits and One Shot competent cells to provide faster, reproducible results to support your downstream applications.

Primary applications

  • Selecting transformed coli colonies using antibiotics
  • Streaking and isolating bacteria cells on LB agar plate
  • Bacteriophage plaque assay using a lawn of bacteria
